Monoclonal mouse anti- human N-terminal proBNP (NT-proBNP), in vitro

Hybridoma clones have been derived from hybridization of Sp2/0 myeloma cells with spleen cells of Balb/c mice immunized with synthetic N-terminal pro brain natriuretic peptide (NT-proBNP), corresponding to amino acid residues 1-12 (MAbs 5B6cc,  29D12cc), a.a.r. 13-27 (MAbs 15F11cc, 13G12cc, 18H5cc, 7B5cc), a.a.r. 22-36 (MAb NT34cc) a.a.r. 28-45 (11D1cc, 16E6cc) and a.a.r. 61-76 (MAbs 24E11cc, 15C4cc). The peptides used for immunization were conjugated with carrier protein.

IgG1 for MAb 18H5cc, NT34cc, 5B6cc, 7B5cc, 11D1cc, 16E6cc

IgG2a for MAbs 29D12cc, 13G12cc, 24E11cc

IgG2b for MAbs 15F11cc, 15C4cc

NT-proBNP and proBNP immunoassay. All MAbs are working in Western Blotting.
Best pairs for sandwich immunoassay (capture-detection): 15F11cc – 24E11cc, 15C4cc – 29D12cc, 15C4cc – 13G12cc, 15C4cc – 18H5cc, 29D12cc-NT34cc. These pairs recognize the antigen in blood of patients with heart failure, unstable angina and myocardial infarction.

MAbs specific to peptides 5-12 (MAb 29D12cc), 13-24 (MAb 15F11cc), 15-20 (MAbs 13G12cc, 18H5cc) and 25-34 (MAb NT34cc) and 15-21 (7B5cc) recognize circulating proBNP and can be used for the development of quantitative proBNP assays in pairs with anti-BNP MAbs (Cat.# 4BNP2). The best pair (capture-detection):
